ABSTRACT
The function of restriction endonucleases is mainly protection against foreign genetic material especially against bacteriophage DNA. The other functions attributed to these enzymes are recombination and transposition. Restriction endonucleases make up the restriction-modification (R-M) systems comprised of endonuclease and methytransferase activities. The endonuclease recognizes and cleaves foreign DNA on the defined recognition sites. The methytransferase modifies the recognition sites in the host DNA and protects it against the activity of endonucleases. The sequences in foreign DNA are generally not methylated and are subjected to restriction digestion. Each restriction enzyme recognizes a specific sequence of 4–8 nucleotides in DNA and cleaves at these sites. According to the description of each reaction in table 1 and table 2; each content was pipetted into the respective labeled tubes which were then centrifuged, incubated at 37°C and then analyzed using agarose gel electrophoresis. After, the molecules were run in agarose gel, the trends for single, double and even triple digestion were observed in L1 through L7 for plasmid DNA, with L1 as molecular weight marker, L6 and L7 as controls.  For mammalian samples, L1-L3 was loaded with sample DNA whereas L4 served as the control. According to the results observed, different endonucleases used in this experiment recognized sequences on both plasmid and mammalian DNA of which they digested with respect to their ability. Problems with enzyme activity can occur under the following conditions: High glycerol concentration, Enzyme-to-DNA ratio is too high; pH is too high, Organic solvents, particularly ethanol and any interference with the DNA molecule. However, by observing the trend of the bands on the results session and the theory behind this experiment, it was therefore concluded that the objectives of the practical were achieved, the activity of the restriction enzymes were demonstrated, digested DNA fragments were observed after they were allowed to run in agarose gel electrophoresis. After digestion, enzymes can be mapped on how and where they can cleave on the DNA sequence.
INTRODUCTION
Restriction endonucleases are enzymes that cleave the sugar-phosphate backbone of DNA. In most practical settings, a given enzyme cuts both strands of duplex DNA within a stretch of just a few bases. Several thousand different restriction endonucleases have been isolated, which collectively exhibit a few hundred different sequence (substrate) specificities. A large majority of restriction enzymes have been isolated from bacteria, where they appear to serve a host-defense role. The idea is that foreign DNA, for example from an infecting virus, will be chopped up and inactivated ("restricted") within the bacterium by the restriction enzyme (Promdonkoy et al, 2003).
Restriction enzymes were discovered in bacteria and there are now more than 1200 known restriction enzyme types. These enzymes are named using a simple system.  EcoRI, for example, was isolated from E. coli and was the first enzyme isolated from a particular strain, hence the designation of I.  HaeIII was isolated from Haemophilus aegyptius. HindIII was isolated from Haemophilus influenzae, and was the third enzyme discovered in a particular strain (Li et al, 1991).

Plasmids are used as vectors to clone DNA in bacteria. One example of a plasmid used for DNA cloning is called pBR322 Plasmid. The pBR322 plasmid contains a gene that allows the bacteria to be resistant to the antibiotics tetracycline and amipicillin. To use pBR322 plasmid to clone a gene, a restriction endonuclease first cleaves the plasmid at a restriction site. pBR322 plasmid contains three restriction sites: PstI, SalI and ecoRI. The first two restriction sites are located within the gene that codes for ampicillin and tetracycline resistance, respectively. Cleaving at either restriction site will inactivate their respective genes and antibiotic resistance. The target DNA is cleaved with a restriction endonuclease at the same restriction site. The target DNA is then annealed to the plasmid using DNA ligase. After the target DNA is incorporated into the plasmid, the host cell is grown in an environment containing ampicillin or tetracycline, depending on which gene was left active. Many copies of the target DNA is created once the host is able to replicate (Jaurez-perez et al, 2002).
Another plasmid used as a vector to clone DNA is called pUC18 plasmid. This plasmid contains a gene that makes the host cell ampicillin resistant. It also contains a gene that allows it to produce beta-galactosidase, which is an enzyme degrades certain sugars. The enzyme produces a blue pigment when exposed to a specific substrate analog. This allows the host to be readily identified. The gene for beta-galactosidase contains a polylinker region that contains several restriction sites. The pUC18 plasmid can be cleaved by several different restriction endonucleases which provide more versatility. When the polylinker sequence is cleaved and the target DNA is introduced and ligased, this inactivates the gene that codes for beta-galactosidase and the enzyme will not be produced. The host cell will not produce a blue pigment when exposed to the substrate analog. This allows the recombinant cells to be readily identified and isolated (Boonserm et al, 2005).
In this practical, the activity of restriction enzymes will be demonstrated by setting up restriction digest using different endonucleases which will either digest the pre-isolated plasmid/mammalian DNA one, two or even three times based on the ability of the enzyme in reading and recognizing the sequences to cleave on the DNA molecule. The digested DNA fragments will further be analyzed using agarose gel electrophoresis (Crickmore et al, 1998).

Reactions tubes were obtained and labeled with respect to the number of reactions to be performed both for table 1 and 2. Following the sequence as in both table 1 and 2 the components of each reaction were added to the respective tubes. By pipetting, each content on each tube were mixed and centrifuged thereafter using a microfuge to collect the contents at the bottom of the tube. The tubes were then incubated at 37 °C for 1 hour. After the completion of the digestion, the technique of gel electrophoresis was used to further analyze the contents on the tubes, this was done by first adding dye to each tube which were kept in -20°C after incubation, then the contents were loaded onto agarose gel and allowed to run
RESULTS
The activity of restriction enzymes were observed after when the sample DNA fragments were allowed to run on an agarose gel electrophoresis following restriction digestion by various enzymes on both plasmid and mammalian DNA which were isolated and purified on the early experiments.

DISCUSSION
Restriction enzymes, also known as restriction endonucleases, are enzymes that cut a DNA molecule at a particular place. They are essential tools for recombinant DNA technology. The enzyme "scans" a DNA molecule, looking for a particular sequence, usually of four to six nucleotides. Once it finds this recognition sequence, it stops and cuts the strands. This is known as enzyme digestion. On double stranded DNA the recognition sequence is on both strands, but runs in opposite directions. This allows the enzyme to cut both strands. Sometimes the cut is blunt; sometimes the cut is uneven with dangling nucleotides on one of the two strands. This uneven cut is known as sticky ends. Restriction enzymes cut at specific sites along the DNA. These sites are determined by the sequence of bases which usually form palindromes. Palindromes are groups of letters that read the same in both the forward and backwards orientation. In the case of DNA the letters are found on both the forward and the reverse strands of the DNA. For example, the 5’ to 3’ strand may have the sequence GAATTC. The complimentary bases on the opposite strand will be CTTAAG, which is the same as reading the first strand backwards! Many enzymes recognize these types of sequences and will attach to the DNA at this site and then cut the strand between two of the bases. After gel analysis of the digested fragments of plasmid DNA, in L1 was observed to have multiple bands since it served as molecular weight marker. L2 and L3 were observed to have linear single bands resulting from a single digest by the restriction enzymes. L4 and L5 had multiple bands resulting from triple digestion, however L5 had RNA contamination as also observed in L6 and L7 which were the controls. By clearly looking at the controls L6 and L7 it clear that the samples were digested by the restriction enzymes, the supercoiled observed in L7 was not observed in any of the samples. Restriction digestions of plasmid DNA are essential for mapping, cloning and other recombinant technologies improving molecular biology. For the mammalian DNA digestion, L1 through L4 only the same kind of band was observed signifying that there was either a single digestion or no digestion at all. beware of the spirit of selfishness and the rebellious spirit, this are unforeseen spirits which have targeted young people, together with the chosen generation of God. these spirits have no sign of humbleness, they seek only things for themselves, they see other people and Christians as demons, they please themselves in adultery and fornication, they crawl in churches like true worshipers while their mission is to destroy the just. they provide leaders with an uncertain kind of pride that have killed many churches, many congregation are still there, many houses of worship are still there, many pastors and young pastors are still there, but dead in the spirit, they are driven by selfishness and rebellion, they think they are prevailing and powerful, but the truth is they are like a little rat inside a sack full of peanuts and only one cat. there is a great reason why the bible says that you should beware of DOGS, beware of evildoers. this spirits can destroy the relationship of a child and a parent, if you are not yet a victim, watch very carefully, if you are already, then consider repenting, don't forget that on earth we live once, and "something that does not end is taboo". Christianity is sharing and loving, and building the Body of Christ into perfection as we wait for His coming! God bless you all....

YOU ARE YOUR OWN DIVINE INSPIRATION! 

The most important thing that many businesses, states, communities, societies, nations, and even individuals are seeking every time they are down is inspiration. Everyone needs to be motivated to get up again; people need ideas that will be fruitful and everything that will keep them moving. A sudden good idea is something that does not just cross the mind at any instance of time, if so; many people would have been rich already since good ideas are worth bringing wealth and divine joy into to one’s life.

Many people consider having mentors and coaches as they live, businesses and state leaderships consider having advisers so to sharpen their capabilities and abilities to successfully run their companies and nations by the inspiration brought by these advisers. However, despite their role and efforts advisers and mentors are playing in your life or your business, it is not in their power to make you do what they say you should do, the power to instigate change always comes from within you, it is you who implement everything.

Sometimes hard times throw themselves upon you and no one is there to inspire you, no one is there to pick you up, to motivate you. People will support you, but not as you will always need and demands it. Every time you fall, don’t be afraid to get up again, to live and love again. Some people are capable of taking you down for no reason; jealousy makes people to do what they end up regretting in the future. Life’s best lessons are often obtained or learned at difficult times, and in those times there is no one to hold your hand and help you up, there is no one to motivate or inspire you to try again. No matter what happens, don’t let hard lessons hardens your heart and your life, don’t be afraid, don’t be shy to get up and try again, yes people will laugh, they will call you names and everything, you know why, because they are afraid that the moment you rise up, you will bring about change, you will take away their business, you will transform everything and everyone around you and they will be ashamed they never helped you.

Mostly you have to go through your worst in order to arrive at your best, when you fall six times, ensure to get up seven times for the perfection of arriving at your destiny, have the spirit of never giving up, the spirit of rising even when you are bleeding tears. The wise always says that tough times never last, but tough people do. You are your own divine inspiration, your own great idea to live your purpose, to live and accomplish your dreams. You don’t need somebody to tell you what to do; you don’t need anyone to drag you to where you want to be. Everything happens for a reason, a failure is not always a mistake, when things happens your opposite way it does not mean that you are rejected, but it means that you are directed and given another chance to do things in a divine way. The mistakes we make today, determines our future tomorrow, don’t forget that you are what you are because of the mistakes you made in the past.

The most inspiration you need and the one that will always change your life for a greater future is yourself. You are the best of you, we always put people into blame even when we first went to them for advises. A person’s advice is sometimes his/her opinion, and everyone’s opinion is not wrong. Be responsible for your own self, your life, and your decisions. Have a great sense of trust in yourself and your ideas don’t wait for people to approve what you think, many people lost their business plans and ideas by doing so. Have a strong trust and believe in yourself, you are your own divine inspiration, the one who will never be wrong, and the one who won’t steal your own ideas. It is your intuition which teaches you that trying times will pass and all your dreams will come true, just be patient.


Live your life, forgive and forget, hope and believe without ceasing or being weary. Whatever you do, always put to your attention that it is you the one you need in order to be the best of you, don’t stop trying, never give up, never stop doing the best of you, a person who never go through hard times has not yet lived, for whatever you are going through today develop you with all the strength and muscles you need to win tomorrow. There is always a point in life where you get tired of chasing everyone and everything they bring into your life and that does not mean you are giving up, but you’ve realized that you don’t need people to tell you who you are, what you are and what you are made for. And that only mean that you are letting go of all kinds of crap that can distract you from being who you are and where you are going. Be the best of you, live your life to the fullest, you are your own inspiration!

The most frequently used English word when people want something from others is love, it is a perfect word used to express how people feel about each other, it is used to outline the liking of something in a great way and or very much. Different people, cultures, societies, denominations and organisation defines and uses the word love in different ways. Love is one precious gift of feeling given to unite friends, families and nations.

The feeling of love is designed in the way that when it is truthfully shared its bonds cannot be broken, it brings about the sense of joy and happiness between the parties. Love is not only limited to sexual feelings, but it has the power to develop the connection between parties and to stimulates success and prosperity to those who hold firmly to each other through love. The feathers and wings of love are so soft better than those of doves; they cover even the deeper parts in which other feelings of liking cannot reach.

Love maintains its value by not being selfish, its integrity by the supernatural powers of caring; it always develop the sense of humor in order to bring joy and happiness to its surroundings; it is not shy to share its deep secrets of harmony and faithfulness, it has strong hopes for a better future, and believes strongly to every positive thoughts for the development of life. Love knows how to bear pains and sufferings; it is patient and does not seek to please only itself. Love is so knowledgeable to an extent of understanding times and seasons, love understands that life is measured by time and that failure to understand times will lead to unforeseen consequences.

It is only love that can bring together great enemies into great friends, that’s another reason you will never find love when you decide to seek it. Love is a precious gift from God, a holy sacrifice that can only be performed by God. It is a great present embedded within the layers of your deep feelings and senses. It is not a magic, but its supernatural powers are so strong greater than any other strong thing you’ve ever known. That’s another reason there is a saying that the best thing you can ever give in this world is LOVE.

The value of love cannot be scored by all the money and minerals that are found on earth, money cannot buy love; it is always stupid for you to charm or be charmed by a dollar. Love is much more valuable than all what money can buy, it has nothing to do with what you do, for wherever it knocks and enter it brings about change, it transforms everyone and everything which accepts it wholeheartedly. Love seals the gaps and wounds which were left by lust and its friends which are there to steal, kill and destroy. Love helps you discover your role in a relationship; it accumulates all the duties you need to configure to ensure a successful relationship and life.

I find it immature when a person claims to love and the other party is convinced that it is love in its truth while it is a raid planned lustfully to destroy him/her. Love is so clear to see, no binoculars or microscopes are needed to tell whether it is love or not. Love’s first intentions are to know and understand the kind of person you are before demanding the extras, love first builds spiritual connections between the two parties before emotional and physical connections comes to take part. Don’t be fooled when a person claims that they love you because of your hot body, the way you dress, talk or walk, true love begins in the spirit, yes you have to feel that different understanding and liking of a partner before you even meet them, the force of attraction first begins in the spirit, the formation of love bonds begins by sharing of spiritual senses just like when atoms share their electrons.

Love is everything, yes everything you need to be happy in this world, love will forever make you happy and maintain your joy. The joy brought by love is far much better than the happiness brought by riches. Love is unique, kind and beautiful, everyone deserves to be loved, and to be loved unconditionally. Therefore, love and don’t lust, keep it and it will keep you safe and sound!
